Question: when people say a peptide has a “short half-life,” are they usually talking about plasma half-life, or functional signalling duration? I keep seeing those used interchangeably and I’m not sure that’s correct.

They’re not the same. Plasma half-life is clearance from blood. Signalling can outlast (or under-last) that depending on receptor kinetics and downstream cascades. Short plasma half-life doesn’t automatically mean short biological effect — and the reverse is also true.

Also watch for assay conditions. Buffer stability studies get quoted as if they’re in vivo half-life. Totally different question.
